Frost was Chairman of the Department of Dermatology, Mt. Sinai Medical Center of Greater Miami, from 1972 to 1990. With Mike Jaharis, the two acquired Key Pharmaceuticals in 1972, ran the company until 1986, selling it to Schering-Plough for $600M. Frost's share of the take was $100M, but most of his wealth derives from the pharmaceutical company he founded, IVAX, a manufacturer of generic drugs (their patents expired), later sold to Israeli firm Teva Pharmaceuticals. He is listed #278 on the list of wealthiest Americans, with an estimated net worth of $1.2B.
